Connective tissue diseases (CTD) are a diverse group of rheumatologic disorders characterized by the presence of autoantibodies and systemic organ involvement, frequently including the lung or chest.

When an autoimmune disorder is suspected the first diagnostic step is the screening for ANAs. These antibodies, directed against components of the cell nucleus and the cytoplasm, were first associated with systemic lupus erythematosus (SLE), but the list of involved diseases has widened because many rheumatic diseases are characterized by the presence of one or more ANAs. In particular, SS-A and SS-B antibodies are associated with SLE and Sjögren's syndrome (SS); dsDNA and Sm antibodies with SLE; RNP antibodies with mixed connective tissue disease (MCTD) and SLE; Scl-70 antibodies with scleroderma; Jo-1 antibodies with polymyositis; centromere antibodies with CREST syndrome.
